We have found this issue to come up occasionally when the customer is running an Anti-Virus software. The anti-virus software can lock the partition and Diskeeper does not have access because the software has already gained exclusive access to the disk at boot up.We advise shutting down your anti-virus temporarily and determine if Diskeeper is able to work.
